چکیده

Soybean lipoxygenases (soybean lipoxygenase-1) has been a well-studied enzyme because it is considered as model for mammalian lipoxygenases. In general, the soybean lipoxygenase-1 used in test of inhibitory activities various compounds. The present study provides bioinformatics approach comparison databases. Their physical and chemical parameters such molecular weight, theoretical pI, amino acid composition, aliphatic index grand average hydropathicity multiple sequence alignments were computed by using several tools. order to see phylogenetic relation among lipoxygenases, tree was constructed. first three most abundant acids 15-lipoxygenase (human) are L (10.3 %), S (7.4 A (6.7 %) (13.3 G V (7.1 respectively. According tree, within separate clade compared conclusion, may not fully characterise human lipoxygenase-15 since there remarkable sequence-based differences, which obtained tools between from other sources especially human. this context, researchers aware problems described above having similar concerns have begun use recombinant DNA technology produce h-15-LOX Escherichia coli.

Soybean leaves contain multiple lipoxygenases.

Chromatofocusing of soybean (Glycine max L.) leaf lipoxygenases revealed three distinct peaks of activity. Based on their isoelectric points (pls), pH optima, and mutant analysis it appears that the leaf isozymes are different from those described from mature soybean seed. At least one leaf lipoxygenase appears to differ from those found in hypocotyls. The pls of the main bands of the three lea...

Structure conservation in lipoxygenases: structural analysis of soybean lipoxygenase-1 and modeling of human lipoxygenases.

Lipoxygenases are a class of non-heme iron dioxygenases which catalyze the hydroperoxidation of fatty acids for the biosynthesis of leukotrienes and lipoxins. The structure of the 839-residue soybean lipoxygenase-1 was used as a template to model human 5-, 12-, and 15-lipoxygenases. A distance-based algorithm for placing side chains in a low homology environment (only the four iron ligands were...

Update on Plant Lipoxygenases Plant Lipoxygenases. Physiological and Molecular Features

Lipoxygenases (LOXs; EC 1.13.11.12) are nonheme iron-containing dioxygenases widely distributed in plants and animals. LOX catalyzes the addition of molecular oxygen to polyunsaturated fatty acids containing a (Z,Z)-1,4-pentadiene system to produce an unsaturated fatty acid hydroperoxide. LOX initiates the synthesis of a group of acyclic or cyclic compounds collectively called oxylipins, which ...

Inhibition of Soybean Lipoxygenases – Structural and Activity Models for the Lipoxygenase Isoenzymes Family

Lipoxygenases (EC 1.13.11.12, linoleate:oxygen, oxidoreductases, LOXs) which are widely found in plants, fungi, and animals, are a large monomeric protein family with non-heme, non-sulphur, iron cofactor containing dioxygenases that catalyze the oxidation of polyunsaturated fatty acids (PUFA) as substrate with at least one 1Z, 4Z-pentadiene moiety such as linoleic, linolenic and arachidonic aci...

The iron ligand sphere geometry of mammalian 15-lipoxygenases.

We investigated the geometry of the iron ligand sphere of the native rabbit 15-lipoxygenase (15-LOX) by X-ray absorption spectroscopy using synchrotron radiation. The soybean LOX-1 was used as a reference compound because its iron ligand sphere is well characterized. For structural information the X-ray absorption spectra were evaluated using the Excurve Program (CCLRC Daresbury Laboratory, War...
